Measuring Media, Water & Yeast for Culturing Fruit Flies

Hi my name is Richard Revis I'm co-owner of Black Jungle Terrarium Supply and on behalf of expertvillage.com we are discussing the culturing of fruit flies. At this point we are ready to start actually setting up the cultures and mixing the media. The first thing that you want to do is make sure you know what the mixture ratio is for the type of media that you are using. The one that we are using today is one third cup dry media to one half cup of water. The first step is to measure out the third cup of media. You don't want to pack the media you just want to shake it down to where it is level and simply pour into the culture container. Some of the medias can be kind of dusty so wearing a dust mask will also help preventing any irritation or allergy's. At this point you would then add the water again making sure that it is level and swirl the culture around to actually insure a even mixture. Most media would appear very thin and runny at this point and that is the appearance that you want. While the media is still wet then take a small pinch of yeast between your fingers and sprinkle in across the surface of the media. Doing while the media is still wet allows the culture to or the moisture of the culture to actually hydrate the yeast and allow it to start growing. After mixing all of the ingredients allow the culture vessel to sit flat for approximately for 1-2 minutes and this allows the culture media to actually solidify. You can see at this stage it is no longer runny. All of the water has been absorb by the media and it is now ready for the addition of the fruit flies.
